General description

Tetraspanin-8 (UniProt P19075; also known as Transmembrane 4 superfamily member 3, Tspan-8, Tumor-associated antigen CO-029) is encoded by the TSPAN8 (also known as TM4SF3) gene (Gene ID 7103) in human. CO-029 is a member of the transmembrane 4 or tetraspanin family of cell-surface 4-transmembrane glycoproteins involved in the regulation of cell activation, growth, and motility. Tetraspanin-8 (CO-029 antigen) was originally characterized as a tumor-associated antigen expressed in gastric, colorectal, and pancreatic cancer cells and exert tumor progression-promoting activity. In metastatic pancreatic and colorectal carcinoma cell lines, PKC activation enhances the colocalization of Tetraspanin-8 and tetraspanin CD151 with integrin α6β4 and subsequent internalization of this integrin-tetraspanin complex, resultinig in decreased cell-matrix adhesion and increased cell migration. Tetraspanin-8 knockdown in HT29 colon cancer cells is reported to significantly reduce cell migratory ability as a result of an upregulation of both integrin-dependent cell-matrix adhesion and calcium-dependent cell-cell adhesion. Human tetraspanin-8 is a four-transmembrane (10-33, 58-72, 84-109, 206-230) protein with both its N- and C-terminal ends (a.a. 1-9 and 231-237) exposed at the cytoplasmic side.
